      标题:肝细胞癌中KIF2C的表达水平及临床意义

      【摘要】 目的 探索KIF2C在肝细胞癌(HCC)组织中的表达及其临床意义。方法 收集唐山市人民医院2009-2013年手术切除的肝细胞癌肿瘤组织及匹配的癌旁组织各104例,采用免疫组化的方法检测肿瘤及癌旁组织中KIF2C的表达水平,并结合Oncomine数据库中的数据对KIF2C在肝癌中的表达情况进行大样本验证。此外,用Kaplan-Meier法分析KIF2C表达高低与患者总生存率(OS)及无瘤生存率(DFS)的关系。结果 HCC组织中KIF2C高表达者占 68.27% (71/104),明显高于癌旁组织的 20.19% (21/104),且差异有统计学意义 (P<0.05);Oncomine数据库中225例HCC及220例癌旁组织标本的KIF2C表达水平验证结果提示,肝癌组织中KIF2C的表达水平明显高于癌旁组织,且差异有统计学意义(P<0.05);KIF2C的表达与患者年龄、肿瘤的大小及肿瘤分化程度以及TNM分期有关(P<0.05);Kaplan-Meier生存分析显示,KIF2C低表达组患者的总生存率和无瘤生存率明显优于KIF2C高表达组。结论 KIF2C在HCC的发生发展过程中可能发挥着重要的作用,KIF2C有望成为HCC治疗的一个潜在靶点,且该基因可以作为HCC患者预后的一个有效的预测因子。

Expression of KIF2C in hepatocellular carcinoma and its clinical significance.

【Abstract】 Objective To investigate the expression of KIF2C in hepatocellular carcinoma (HCC) and its clini-cal significance. Methods A total of 104 cases of hepatocellular carcinoma tissues and matched para-cancer tissueswere collected from Tangshan People's Hospital from 2009 to 2013. The expression of KIF2C in HCC tissues and adja-cent normal tissues was detected by immunohistochemistry, and the prognosis of patients was verified with the data inthe Oncomine database. Furthermore, Kaplan-Meier analysis was used to evaluate overall survival (OS) and disease-freesurvival (DFS) of HCC patients. Results The high KIF2C expression of HCC tissue accounted for 68.27% (71/104),which was significantly higher than 20.19% (21/104) of adjacent tissues, and the difference was statistically significant(P<0.05). The verification results of KIF2C expression in 225 cases of HCC and 220 paraneoplastic tissues in the On-comine database suggested that the expression level of KIF2C in HCC tissues was significantly higher than that in theadjacent tissues (P<0.05). The expression of KIF2C was associated with the age of the patients, the size of the tumor,the degree of tumor differentiation, and the TNM stage (P<0.05). Kaplan-Meier analysis revealed that the overall sur-vival rate of patients with low KIF2C expression was significantly better than that of individuals with high expression.Conclusion KIF2C may play an important role in the development and progression of HCC. KIF2C is expected to bea potential target for HCC therapy and this gene may serve as a valid predictor of prognosis in patients with HCC.
