      标题:兔膝关节海水浸泡损伤的实验研究

      【摘要】 目的 探索兔膝关节海水浸泡损伤关节液炎性因子的特点及关节软骨的组织学改变。方法 取40只新西兰白兔,建立兔膝关节开放性损伤模型,随机分为实验组和对照组,每组 20只。实验组海水浸泡膝关节3 h;对照组生理盐水浸泡膝关节3 h。术后3 d,抽取全部实验动物患膝关节液,采用酶联免疫吸附法检测关节液中白细胞介素 1β (IL-1β)和肿瘤坏死因子α (TNF-α)的含量。术后 7 d取兔膝关节股骨髁部组织,行HE染色,观察关节软骨的组织学改变。结果 术后 3 d,实验组白兔的膝关节液 IL-1β和TNF-α水平分别为(34.72±5.79) pg/mL和(321.13±38.44) pg/mL,明显高于对照组的(25.48±3.13) pg/mL和(248.25±48.10) pg/mL,差异均有统计学意义(P<0.05);术后7 d,实验组白兔的髁部软骨层细胞排列紊乱,局部潮线消失,可见增生的纤维组织;对照组关节软骨组织结构无明显改变。结论 兔膝关节海水浸泡损伤模型在损伤后3 d可见由 IL-1β和TNF-α介导的炎性反应,损伤后7 d关节软骨的组织学结构破坏明显。

Study on the seawater immersion of knee injury in a rabbit model.

【Abstract】 Objective To observe the characteristics of inflammatory factors in the synovial fluid and the his-tology of the cartilage in a rabbit model undergoing seawater immersion after knee injury. Methods Forty rabbit mod-els of knee injury were randomly divided into two groups: control group (n=20) and experimental group (n=20), whichreceived seawater immersion and normal saline immersion for 3 hours, respectively. After 3 days, the content of interleu-kin-1 β (IL-1 β) and tumor necrosis factor alpha (TNF-α) in the synovial fluid in each group was measured by en-zyme-linked immunosorbent assay. After 7 days, the histological change in cartilage of knee was observed by HE staining.Results After 3 days, the IL-1β and TNF-α levels in synovial fluid were (34.72±5.79) pg/mL and (321.13±38.44) pg/mLin experimental group, versus (25.48±3.13) pg/mL and (248.25±48.10) pg/mL in control group (P<0.05). After 7 days,the cartilage in experimental group showed irregular structure, with the cartilage cells arranged irregularly and prolifera-tion of fibrous tissue observed, while the cartilage in the control group showed no obvious change. Conclusion In therabbit model of seawater immersion after knee injury, inflammatory reaction mediated by IL-1β and TNF-α in the syno-vial fluid was observed, and histological structure in the cartilage of knee is destroyed obviously.
