      标题:ADAMTS10在心肌梗死后心肌纤维中的表达及其意义

      【摘要】 目的 观察ADAMTS10在大鼠心肌梗死后心肌纤维中的表达,并分析其在心肌纤维化中起作用的方式及意义。方法 24只8周龄雄性SD大鼠随机分为两组各12只,心肌梗死模型组采用冠状动脉左前降支结扎方法构建,假手术组只缝针不结扎冠状动脉。造模4周后超声心动图检测心功能,天狼星红染色检测心肌纤维化程度及胶原容积分数,实时荧光定量 PCR方法检测 ADAMTS10、fibrillin-1 mRNA表达,Western-blot方法检测ADAMTS10蛋白表达。结果 与假手术组相比,心肌梗死组大鼠心室明显扩张[左心室收缩末期内径(LVESD):(4.63±0.39) mm vs (1.98±0.26) mm,左心室舒张末期内径(LVEDD):(5.79±0.37) mm vs (3.37±0.31) mm],差异均有显著统计学意义(P<0.01);心功能严重受损[左心室射血分数(EF):(81.20±4.75)% vs (31.02±3.61)%,短轴缩短率(FS):(38.91±3.53)% vs (17.50±1.94)%],差异均有显著统计学意义(P<0.01);心肌组织纤维化程度加重,心肌胶原容积分数增加[(12±0.14)% vs (2±0.19)%],差异有显著统计学意义(P<0.01);心肌梗死大鼠心肌组织中ADAMTS10 mRNA表达水平[(4.53±0.07) vs (1.00±0.03)]及蛋白表达水平[(1.87±0.02) vs (1±0.14)]均升高,且 fibrinllin-1 mRNA在心肌梗死大鼠心肌组织中表达也明显升高[(3.88±0.13) vs(1±0.02)],差异均有显著统计学意义(P<0.01)。结论 ADAMTS10在梗死后心肌纤维化过程中发挥着重要作用,其可能通过调控纤维蛋白 1的表达参与心肌纤维化的过程;ADAMTS10可能成为心肌纤维化诊断和治疗靶点。

Expression and significance of ADAMTS10 in myocardial fibrosis after acute myocardial infarction in rats.

【Abstract】 Objective To investigate the expression level of ADAMTS10 in myocardial fibrosis after acutemyocardial infarction, and to analyze its mechanism and significance in myocardial fibrosis. Methods A total of 24male SD rats aged 8 weeks were randomly divided into the sham group (n=12, the coronary artery was sutured, but not li-gated) and myocardial infarction model group (n=12, left anterior descending branch of coronary artery was ligated).Four weeks after the ligation, echocardiography was used to detect cardiac function, and myocardial fibrosis degree andthe collagen volume fraction were detected by Sirius red staining. Real-time PCR was used to detect mRNA expressionof ADAMTS10 and fibrillin 1. Western blot was applied to detect the protein expression of ADAMTS10. Results Com-pared with sham group, rats in myocardial infarction model group displayed greater ventricular expansion [left ventricu-lar end systolic diameter (LVESD) (4.63±0.39) mm vs (1.98±0.26) mm, left ventricular end diastolic diameter (LVEDD)(5.79±0.37) mm vs (3.37±0.31) mm, P<0.01], seriously damaged heart function [ejection fraction (EF) (81.20±4.75)% vs(31.02±3.61)%, fractional shortening (FS) (38.91±3.53)% vs (17.50±1.94)%, P<0.01], aggravated myocardial tissue fi-brosis degree, and increased myocardial collagen volume fraction [(12±0.14)% vs (2±0.19)%, P<0.01]. Moreover, the ex-pression of ADAMTS10 mRNA [(4.53±0.07) vs (1.00±0.03), P<0.01) and protein levels [(1.87±0.02) vs (1±0.14), P<0.05] were elevated, and fibrinllin-1 mRNA expression in myocardial tissue of rats with myocardial infarction was alsoup-regulated [(3.88 ±0.13) vs (1 ±0.02), P<0.01]. Conclusion ADAMTS10 plays an important role in the process ofmyocardial fibrosis after myocardial infarction, and it may be involved in the process of myocardial fibrosis by regulat-ing the expression of fibrillin-1. ADAMTS10 may be one of the diagnosis and therapeutic targets of myocardial fibrosisin the future.
