      标题:miR-21、FOXO3a、STAT3表达与胃癌及临床病理参数的关系

      【摘要】 目的 探究miR-21、FOXO3a、STAT3表达与胃癌及临床病理参数的关系。方法 收集2015年1月至2016年6月菏泽市立医院病理科和菏泽医学专科附属医院病理科首发胃癌手术切除患者癌及相应癌旁胃组织标本共80例。应用实时荧光定量PCR (qRT-PCR)检测组织中miR-21、FOXO3a、STAT3表达情况,应用免疫组织化学析组织中FOXO3a和STAT3蛋白表达情况。结果 与癌旁胃组织比较,胃癌组织中miR-21表达水平显著增加,FOXO3a mRNA表达水平显著降低,STAT3 mRNA表达水平显著增加,差异均有统计学意义(P<0.05)。免疫组织化学显示,与癌旁胃组织比较,癌组织中FOXO3a显示弱阳性或阴性表达,而STAT3蛋白显示强阳性表达。随着癌组织在胃壁中浸润程度的增加、淋巴结转移的出现,miR-21和STAT3 mRNA表达水平逐渐增加,而FOXO3a表达水平逐渐降低,差异均有统计学意义(P<0.05)。在胃癌组织中,miR-21表达水平与FOXO3a mRNQA表达水平呈负相关(P<0.05),miR-21表达水平与STAT3 mRNA表达水平呈正相关(P<0.05),FOXO3a与STAT3 mRNA表达水平呈负相关(P<0.05)。结论 miR-21、STAT3、FOXO3a在胃癌中的差异性表达与胃癌患者癌组织浸润、淋巴结转移等临床病理特征密切相关。

Expression of miR-21, FOXO3a and STAT3 in gastric carcinoma and their relationship with clinicopathologicalparameters.

【Abstract】 Objective To explore the expression of miR-21, FOXO3a, STAT3 in gastric carcinoma and theirrelationship with clinical pathological parameters. Methods From January 2015 to June 2016, 80 samples of cancerand corresponding adjacent gastric tissues were collected from the gastric cancer patients undergoing gastrectomy in theHeze city Hospital Affiliated Hospital of Heze Medical College. The expression of miR-21, FOXO3a and STAT3 weredetected by real-time quantitative PCR (qRT-PCR). Expression of FOXO3a and STAT3 protein in tissues were detectedby immunohistochemical analysis. Results Compared with the adjacent gastric tissue, the expression of miR-21 in gas-tric carcinoma was significantly increased, the expression of FOXO3a mRNA was significantly decreased, and the ex-pression of STAT3 mRNA was significantly increased (P<0.05). Immunohistochemistry indicated that FOXO3a showedweakly positive or negative expression compared with adjacent gastric tissue, while STAT3 protein showed strong posi-tive expression. With the emergence of the increasing degree of tumor invasion and lymph node metastasis in gastric tis-sue, the expression of miR-21 and STAT3 mRNA increased gradually, while the expression level of FOXO3a decreased(P<0.05). In gastric cancer tissues, the expression of miR-21 was negatively correlated with FOXO3a and mRNQA ex-pression (P<0.05); the expression of miR-21 was positively correlated with STAT3 and mRNA (P<0.05); and the expres-sion of FOXO3a was negatively correlated with STAT3 mRNA (P<0.05). Conclusion The differential expression ofmiR-21, STAT3 and FOXO3a in gastric carcinoma is closely related to the clinicopathological features of invasion andlymph node metastasis in gastric cancer patients.
