      标题:淫羊藿苷对人骨关节炎软骨细胞增殖凋亡的影响

      【摘要】 目的 研究淫羊藿苷对人骨关节炎软骨细胞增殖凋亡的影响。方法 体外分离培养人骨关节炎软骨细胞,用不同浓度的淫羊藿苷作用于人骨关节炎软骨细胞,噻唑蓝(MTT)法检测细胞增殖,流式细胞术检测细胞凋亡,酶联免疫吸附测定法(ELISA)检测细胞培养液上清中白细胞介素-1β (IL-1β)水平,Western blot检测细胞中基质金属蛋白酶-1 (MMP-1)、基质金属蛋白酶-3 (MMP-3)、基质金属蛋白酶-13 (MMP-13)的表达。结果 0、50 μg/mL、100 μg/mL、150 μg/mL的淫羊藿苷作用后的骨关节炎软骨细胞OD值依次为(0.58±0.04)、(0.76±0.06)、(0.92±0.07)、(1.26±0.09),凋亡率依次为(9.35±0.27)%、(8.69±0.11)%、(7.41±0.12)%、(6.87±0.09)%,细胞分泌的IL-1β依次为(10.53±0.97) pg/mL、(7.94±0.84) pg/mL、(6.05±0.71) pg/mL、(5.02±0.49) pg/mL,MMP-1水平依次为(0.25±0.02)、(0.20±0.01)、(0.16±0.01)、(0.11±0.02),MMP-3水平依次为(0.11±0.01)、(0.08±0.01)、(0.05±0.01)、(0.02±0.01),MMP-13水平依次为(0.39±0.03)、(0.30±0.02)、(0.21±0.01)、(0.19±0.01)。以上各项指标比较,差异均有统计学意义(P<0.05)。结论 淫羊藿苷能够促进人骨关节炎软骨细胞增殖,抑制人骨关节炎软骨细胞凋亡,作用机制可能与细胞分泌的 IL-1β和细胞中MMP-1、MMP-3、MMP-13的表达水平有关。

Effects of icariin on proliferation and apoptosis of human osteoarthritis chondrocytes.

【Abstract】 Objective To study the effects of icariin on the proliferation and apoptosis of human osteoarthritischondrocytes. Methods Human osteoarthritis chondrocytes were isolated and cultured in vitro, and then treated withdifferent concentrations of icariin. Cell proliferation was detected by MTT assay, and cell apoptosis was detected by flowcytometry. Interleukin-1beta (IL-1β) in supernatant of cell culture was detected by ELISA, and western blot was used todetect the expression of matrix metalloproteinase-1 (MMP-1), MMP-3 and MMP-13 in the cells. Results After treat-ment with 0 μg/mL, 50 μg/mL, 100 μg/mL and 150 μg/mL icariin, the OD values were (0.58±0.04), (0.76±0.06), (0.92±0.07), (1.26±0.09); the rate of apoptosis were (9.35±0.27)%, (8.69±0.11)%, (7.41±0.12)%, (6.87±0.09)%; the secretionlevels of IL-1β were (10.53±0.97) pg/mL, (7.94±0.84) pg/mL, (6.05±0.71) pg/mL, (5.02±0.49) pg/mL; MMP-1 expres-sion levels were (0.25±0.02), (0.20±0.01), (0.16±0.01), (0.11±0.02); MMP-3 expression levels were (0.11±0.01), (0.08±0.01), (0.05±0.01), (0.02±0.01); MMP-13 expression levels were (0.39±0.03), (0.30±0.02), (0.21±0.01), (0.19±0.01).There were statistically significant differences in the above indexes (P<0.05). Conclusion Icariin can promote the pro-liferation and inhibit the apoptosis of human osteoarthritis chondrocytes. The mechanism might involve the expressionof MMP-1, MMP-3 and MMP-13 and the levels of IL-1β secreted by the cells.
