      标题:枇杷清痤胶囊对丙酸杆菌致大鼠痤疮模型的实验研究

      【摘要】 目的 建立大鼠耳廓痤疮模型,观察枇杷清痤胶囊对大鼠耳廓痤疮模型的治疗效果,探讨其作用机制。方法 SD大鼠60只,按体重均衡随机分为正常对照组、模型对照组、阳性对照组(金花消痤丸,1.08 g/kg)及枇杷清痤胶囊高(1.296 g/kg)、中(0.648 g/kg)、低(0.324 g/kg)剂量组,每组10只。大鼠右耳耳廓皮内注射痤疮丙酸杆菌液制造大鼠耳廓痤疮模型,造模成功后,各组分别灌胃给药,每日1次,连续7 d,末次给药后,测定大鼠耳廓厚度、血清中白细胞介素-1α (IL-1α)、可溶性白细胞介素-2受体(sIL-2R)和睾酮含量。结果 与模型对照组比较,枇杷清痤胶囊高、中剂量组和阳性对照组能显著降低大鼠耳廓肿胀度,差异有显著统计学意义(P<0.01);与模型对照组比较,枇杷清痤胶囊高剂量组能显著降低血清中 IL-1α含量(P<0.01),枇杷清痤胶囊中、低剂量组及阳性对照组也能降低血清中 IL-1α含量,差异有统计学意义(P<0.05);与正常对照组比较,模型对照组血清中 sIL-2R和睾酮含量无明显变化,差异无统计学意义(P>0.05),说明造模不会影响大鼠血清中 sIL-2R和睾酮含量。结论 枇杷清痤胶囊对大鼠耳廓痤疮模型所致炎症有较好的治疗作用,其作用机制可能与抑制和减少 IL-1α的产生有关。

Effect of Pipa Qingcuo Capsule on Propionibacterium acnes-induced rat model with acne on the auricle.

【Abstract】 Objective To establish a rat model of acne on the auricle and evaluate the therapeutic effect of PipaQingcuo capsule and its related mechanism of treating acne. Methods Sixty SD rats were randomly divided into nor-mal control group, model control group, positive control group (Jihua Xiaocuo Pill, 1.08 g/kg) and groups treated withPipa Qingcuo Capsule of high (1.296 g/kg), medium (0.648 g/kg) and low (0.324 g/kg) doses, with 10 rats in each group.With the establishment of acne model by intradermal injection of Propionibacterium acnes into inner auricles of rats,each group were administered orally, 1 time daily for 7 consecutive days. After the last delivery, rat auricle thickness, se-rum interleukin-1 alpha (IL-1α) and soluble interleukin-2 receptor (sIL-2R) content and testosterone levels were deter-mined. Results Compared with the model control group, Pipa Qingcuo Capsule group of high and medium doses andpositive control group could significantly reduce the rat auricle swelling degree (P<0.01). Compared with the model con-trol group, Pipa Qingcuo Capsule group of high, medium and low doses, positive control group could all reduce the con-tent of serum IL-1α significantly (P<0.01 for high dose, and P<0.05 for others). Compared with the normal controlgroup, model control group showed no significant changes in serum sIL-2R content and testosterone level (P>0.05), indi-cating that model control group did not affect serum sIL-2R content and testosterone level. Conclusion Pipa QingcuoCapsule provides good therapeutic effect on inflammation caused by rat model with acne on the auricle, and the mecha-nism of treating acne might involve inhibiting and reducing the content of IL-1α.
