      标题:香菇多糖对肿瘤坏死因子-α诱导炎症细胞模型的作用

      【摘要】 目的 研究香菇多糖对TNF-α诱导炎症细胞模型的作用。方法 选取小鼠RAW264.7巨噬细胞为研究载体,实验分为空白组、模型组、香菇多糖高、中和低浓度组,共5组,每组设5个复孔。采用1 g/L、10 g/L、100 g/L TNF-α诱导建立炎症细胞模型,予香菇多糖进行给药孵育,于37℃及5% CO2的条件下培养6 h,取上清样品备测 IL-6、IL-18与IL-23。采用酶联免疫法检测 IL-6、IL-18与 IL-23含量水平。结果 采用10 g/L TNF-α诱导成功建立炎症细胞模型。药物孵育后,5组的 IL-6、IL-18及 IL-23水平比较差异均有统计学意义(P<0.05)。其中,香菇多糖高、中、低浓度组和模型组的 IL-6 [(38.85±4.49) pg/mL、(56.53±7.41) pg/mL、(82.16±5.48) pg/mL vs (97.50±12.04) pg/mL]、IL-18[(28.26±4.54) pg/mL、(43.71±5.29) pg/mL、(52.62±6.55) pg/mL vs (80.27±6.90) pg/mL]及 IL-23 [(60.63±4.02) pg/mL、(74.91±7.64) pg/mL、(90.51±8.73) pg/mL vs (121.31±9.97) pg/mL]水平比较,香菇多糖高、中、低浓度组明显低于模型组,差异均有统计学意义(P<0.05)。香菇多糖高浓度组的 IL-18水平与空白组相当[(28.26±4.54) pg/mL vs (17.76±1.22) pg/mL],差异无统计学意义(P>0.05)。结论 香菇多糖能够抑制TNF-α诱导炎症细胞模型的 IL-6、IL-18和IL-23,故而具有免疫调节作用,且高浓度香菇多糖能够显著减少 IL-18的分泌。

【Abstract】 Objective To investigate the effect of lentinan in treating the cell model of inflammatory responseinduced by tumor necrosis factor (TNF)-α. Methods RAW 264.7 cell was used as the study model, and there were 5groups including the blank group, model group, high, medium and low concentrations of lentinan group. The inflammato-ry cell models induced by TNF-α (1 g/L, 10 g/L, 100 g/L). Then under the situation of 37℃ and 5% CO2, test groupswere added lentinan to have the incubation with 6 h, and the supernatant was collected. The concentrations of interleukin(IL)-6, IL-18 and IL-23 were tested by enzyme-linked immunosorbent assay (ELISA). Results It is successful to use10 g/L TNF-α to induce the inflammatory cell model.
