      【摘要】 目的 观察乙型肝炎病毒(HBV)对SOX4表达的影响，并探讨其机制。方法 RT-PCR和Western印迹法检测HepG2和HepG2.2.15细胞中 SOX4 mRNA和蛋白表达的差异，将HBV感染性克隆 pHBV1.3及其空载体分别与含 SOX4基因启动子的报告质粒 pGL3-SOX4共转染HepG2细胞，测定荧光色素酶的活性，RT-PCR和Western印迹法分别检测 SOX4 mRNA和蛋白表达的情况。结果 HepG2.2.15细胞中 SOX4 mRNA和蛋白的表达水平明显高于HepG2，HBV能够激活 SOX4基因启动子的活性，并在mRNA和蛋白水平上调 SOX4的表达。结论 HBV能够在转录和翻译水平上调SOX4的表达。

【Abstract】 Objective To investigate the effect of hepatitis B virus (HBV) on the expression of SOX4 and itsregulatory mechanism. Methods The expression of SOX4 mRNA and protein in HepG2 and HepG2.2.15 cells was mea-sured by RT-PCR and Western blot. HBV infectious clone pHBV1.3 and its empty vector were co-transfected intoHepG2 cells with reporter plasmid pGL3-SOX4 containing SOX4 gene promoter. The luciferase activity was measured,and expression of SOX4 mRNA and protein were measured by RT-PCR and Western blot. Results The expression ofSOX4 mRNA and protein was higher in HepG2.2.15 cells than in HepG2 cells. HBV could activate the activity of SOX4promoter, and the expression of SOX4 were upregulated at mRNA and protein level. Conclusion HBV can upregulatethe expression of SOX4 both at the transcription and translation level.
