      标题:大蒜素联合5-FU对腺样囊性癌ACC-M裸鼠移植瘤的抑制作用

      【摘要】 目的 探讨大蒜素联合5-FU对腺样囊性癌ACC-M裸鼠移植瘤的抑制作用。方法 建立ACC-M细胞Balb/c裸鼠肩胛下移植瘤模型30只。造模2周后挑选24只肿瘤大小较均匀的裸鼠随机分为实验组(大蒜素组、5-FU组和大蒜素联合5-FU组)和对照组,各6只。实验组按照体质量 0.01 mL/g腹腔注射大蒜素和5-FU浓度均为20 μg/mL,对照组注射0.9% PBS液,每两天一次,共3次,末次注射1周后处死动物。测量移植瘤的体积变化,HE染色观察移植瘤的病理学改变。结果 移植瘤体积变化比较结果:各实验组移植瘤体积明显缩小,5-FU组体积小于大蒜素组,大蒜素联合 5-FU组与大蒜素组、5-FU组比较差异有显著统计学意义(P<0.01);镜下见对照组的癌细胞生长旺盛,异型率高,而各实验组的肿瘤细胞出现不同程度减少,间质增加,大蒜素联合 5-FU组最为明显。结论 大蒜素和5-FU均能有效抑制ACC-M细胞Balb/c裸鼠移植瘤的生长,且大蒜素联合5-FU的抑制作用明显强于大蒜素或5-FU单独使用。

Inhibitory effects of allicin combined with 5-FU on adenoid cystic carcinoma ACC-M in nude mice model.

【Abstract】 Objective To investigate the inhibitory effect of allicin combined with 5-fluorouracil (5-FU) onadenoid cystic carcinoma in nude mice models transplanted with ACC-M cells. Methods The nude mice xenograftmodels were established by subcutaneous injecting human adenoid cystic carcinoma cell line ACC-M into Balb/c nudemice. Twenty-four xenograft model rats were randomly divided into four groups (control group and three experimentalgroups of allicin group, 5-FU group, and allicin combined with 5-FU group), with 6 rats in each group. The rates in thefour groups received intraperitoneal injection of 0.01 mL/g medicine according to the weight of each nude mice, 0.9%phosphate buffer saline (PBS), 20 μg/mL allicin, 20 μg/mL 5-FU, and 20 μg/mL allicin combined with 20 μg/mL 5-FUrespectively, once per two days, for three times. The animals were executed after 1 week for measuring the xenograft tu-mor volume and observing the pathological changing by HE staining. Results The xenograft tumor volume of three ex-perimental groups had narrowed considerably. The tumor volume was significantly decreased in the 5-FU group com-pared with Allicin group, with statistically significant difference between allicin combined with 5-FU group and allicingroup, 5-FU group (P<0.01). Under microscope, the cancer cells of the control group showed vigorous growth and highabnormal rate, while the cancer cells of the three experimental groups showed reduced quantity and increased intercellu-lar substance to different extent, especially in allicin combined with 5-FU group. Conclusion Allicin and 5-FU canboth effectively inhibit the growth of nude mice xenograft tumor of adenoid cystic carcinoma. And the inhibitory effectsof allicin combined with 5-FU is more significant than either allicin or 5-FU used alone.
