      【摘要】 目的 分析竹叶柴胡不同部位总黄酮含量,研究黄酮在柴胡不同部位的分布,为中药资源合理开发利用
提供参考。方法 以芦丁为对照品,以5%硝酸铝与样品中黄酮成分产生络合反应,选择激发波长λex=445 nm,发射波
长λem=484 nm,应用荧光光度法测样品的荧光光度值,计算总黄酮含量。结果 以芦丁在0.198~3.168 μg/mL范围
内与荧光强度线性关系良好,y=0.972 8x-0.001,r2=0.999 3。竹叶柴胡花、叶、茎中总黄酮含量最分别为 3.08%、
1.85%、0.41%。结论 竹叶柴胡花、叶中总黄酮含量较高,应在开花期采收竹叶柴胡地上部分。

Determination of total flavonoids in different aerial parts of Bupleurum marginatum by fluorescence
spectrophotometry.

【Abstract】 Objective To establish a method to determine the total flavonoid levels in flower, leaves and stalk
of Bupleurum marginatum by Spectrofluorimetry, and to provided a reference for reasonable development and utilization
of traditional Chinese medicine resources. Methods Based on fluorescent property that flavonoids could make a stable
complexation with Al3+, and the excitation and emission wavelengths were selected at 445 nm and 484 nm respectively
with rutin as the standard sample. The total flavonoids levels were determined in the sample by fluorescence spectropho-
tometry. Results The fluorescence intensity had a good linear relationship with rutin concentration in the range of
0.198~3.168 μg/mL. The regression equation was found to be y=0.972 8x-0.001, r2=0.999 3. The content of flavonoids
in flower, leaves and stalk were 3.08%, 1.85%, 0.41%, respectively. Conclusion The flower, leaves of Bupleurum mar-
ginatum contain relatively higher levels of total flavonoids, which provides a reference for harvesting the aerial parts of
Bupleurum marginatum in flowering period.
