      标题:TNFSF4基因rs3850641多态性与2型糖尿病的相关性研究

      【摘要】 目的 探讨四川南充地区汉族人 TNFSF4基因 rs3850641多态性与 2型糖尿病(T2DM)的相关性。
方法 应用聚合酶链反应-限制性片段长度多态性技术检测于2014年7月至2015年6月选取的190例正常对照者
和218例T2DM患者(T2DM组)的 rs3850641多态性。葡萄糖氧化酶法测定两组受检者的血糖(Glu)水平,氧化酶法
测定两组受检者的血清甘油三酯(TG)、总胆固醇(TC)及高密度脂蛋白胆固醇(HDL-C)水平。结果 TNFSF4基因
rs3850641位点等位基因频率和基因型频率分布均符合Hardy-Weinberg平衡定律;rs3850641多态位点G等位基因
频率为 0.161;T2DM组和对照组之间 rs3850641位点等位基因的频率差异无统计学意义[0.163 (71/168) vs 0.158
(60/190),P>0.05];对照组和T2DM组 rs3850641位点G基因型携带者的血糖水平分别高于各组AA基因型携带者的
血糖水平[(4.72±1.43) mmol/L vs (4.50±1.30) mmol/L、(6.44±2.45) mmol/L vs (6.23±2.16) mmol/L],差异均有统计学意
义(P<0.05)。结论 TNFSF4基因 rs3850641多态性与T2DM无关联,但其G等位基因与血糖水平相关。

Analysis of TNFSF4 gene rs3850641 polymorphisms in patients with type 2 diabetes mellitus.

【Abstract】 Objective To investigate the variations of TNFSF4 gene rs3850641 polymorphisms and their relation-
ships with type 2 diabetes mellitus (T2DM) among people with Han nationality in Nanchong area of Sichuan Province.
Methods The genotypes and allele frequencies of rs3850641 polymorphism in 218 T2DM patients and 190 control sub-
jects, who were selected from July 2014 to June 2015, were assayed by polymerase chain reaction and restriction fragment
length polymorphism (PCR-RFLP). Serum glucose and lipids of the two groups were measured respectively by glucose
oxidase method and oxidase enzymatic kits. Results TNFSF4 gene rs3850641 allele and genotype distributions were in
Hardy-Weinberg equilibrium. The frequencies of G allele at rs3850641 site were 0.161 in total. There was no significant
difference of allele and genotype frequencies between T2DM and control groups, 0.163 (71/168) vs 0.158 (60/190), P>
0.05. Subjects with genotype GG and AG at rs3850641 sites had a significantly higher serum glucose than those with
genotype AA in both control and T2DM groups, (4.72 ± 1.43) mmol/L vs (4.5 ± 1.30) mmol/L, (6.44 ± 2.45) mmol/L vs
(6.23±2.16) mmol/L, P<0.05. Conclusion These results suggest that rs3850641 polymorphism in the TNFSF4 gene
may has an effect on serum glucose, but not with type 2 diabetes mellitus in the population group.
