      标题:AQP1、AQP5、AQP8和mRNA在羊水量异常患者中的表达与意义

      【摘要】 目的 探讨水通道蛋白1 (AQP1)、水通道蛋白5 (AQP5)、水通道蛋白8 (AQP8)与羊水量调节的相关
性,以期阐明羊水量异常的分子细胞病理学机制。方法 收集我院2012年1月至2015年12月行剖宫产分娩的产
妇120例,其中特发性羊水量过少组40例、过多组40例和羊水量正常组40例,采用Western blotting和Q-PCR检测
AQP1、AQP5、AQP8蛋白和mRNA的表达,统计分析与其羊水异常的相关性。结果 相对于正常组,羊水过少
组中AQP1的mRNA表达为下调,但AQP1的蛋白表达却为上调,反之在羊水过多组中AQP1的mRNA为上调,
蛋白的表达却为下调,其组间差异具有统计学意义(P<0.05);羊水过多组与过少组中AQP5的mRNA表达均为下
调,且蛋白表达均为下调,其组间差异具有统计学意义(P<0.05);羊水过少组AQP8的mRNA表达均为下调,蛋白
表达却为上调,但羊水过多组中AQP8的mRNA为上调,蛋白的表达却为下调,其组间差异具有统计学意义(P<
0.05)。结论 AQP1和AQP8蛋白的低表达可能会造成羊水吸收减少而导致羊水过多,AQP5蛋白的表达下调可
能会引起羊水代谢的异常而导致羊水量异常。

Expression and significance of AQP1, AQP5, AQP8 and mRNA in patients with abnormal amniotic fluid volume.

【Abstract】 Objective To investigate the correlation between AQP1, AQP5, AQP8 and amniotic fluid volume
regulation, in order to further elucidate the molecular mechanism of abnormal amniotic fluid volume. Methods A total
of 120 cases of lying-in woman underwent cesarean section, who admitted to our hospital from January 2012 to Decem-
ber 2015, were selected as the research objects and divided into the oligohydramnios group, polyhydramnios group and
normal group, with 40 cases in each group. AQP1, AQP5, AQP8 mRNA and protein expression levels were detected by
Western blotting and Q-PCR, and the correlation with abnormal amniotic fluid volume was analyzed. Results Com-
pared with the normal group, AQP1 mRNA level in the oligohydramnios group was down-regulated, and AQP1 protein
expression was up-regulated, whereas AQP1 mRNA was up-regulated and protein expression was down-regulated in the
polyhydramnios group. The differences between the groups were statistically significant (P<0.05). AQP5 mRNA level
and protein expression were down-regulated in the oligohydramnios group, polyhydramnios group, and the difference be-
tween the groups was statistically significant (P<0.05). In the oligohydramnios group, AQP8 mRNA was down-regulated
and AQP8 protein expression was up-regulated, whereas in the oligohydramnios group, AQP8 mRNA was up-regulated
and AQP8 protein expression was down-regulated. The differences between the groups were statistically significant (P<
0.05). Conclusion The low protein expression of AQP1 and AQP8 may result in a reduction of amniotic fluid absorp-
tion, resulting in polyhydramnios. The down-regulated protein expression of AQP5 may cause the abnormal amniotic flu-
id metabolism, resulting in too much or too little amniotic fluid volume.
