      【摘要】 目的 观察血管生成抑制素治疗血管瘤的效果。方法 发酵毕赤酵母工程菌组成型表达重组血
管生成抑制素,用SP-Sepharose Fast Flow (阳离子交换剂)柱进行蛋白纯化;通过抑制鸡胚绒毛尿囊膜(CAM)血管
生成试验和诱发血管内皮细胞凋亡试验,分析重组血管生成抑制素的生物学活性,在血管瘤模型(公鸡肉垂)中注
射血管生成抑制素,分析血管生成抑制素对血管瘤的药效。结果 血管生成抑制素在毕赤酵母中高效表达,经
过纯化,收获达98%的重组血管生成抑制素;重组血管生成抑制素具有抑制CAM血管生成和诱发血管内皮细胞凋
亡的活性;重组血管生成抑制素与血管瘤模型作用14 d后,瘤体血管生长抑制率为51% (P<0.01),血管瘤增殖抑制
率为39% (P<0.05)。结论 成功制备重组血管生成抑制素药物,血管生成抑制素对于血管瘤模型具有显著的药效。

Study on treating angeioma by angiostatin.

【Abstract】 Objective To observe the effect of using angiostatin to treat angeioma. Methods Recombinant
angiostatin was constitutively expressed in Pichia pastoris by fermentation and then purified by SP-Sepharose Fast
Flow (cation exchanger). Biological activity of the recombinant angiostatin was analyzed by the tests of suppression
angiogenesis of chorioallantoic membrane of chick embry (CAM) and inducing apoptosis of vascular endothelial
cells. The drug effect for angeioma was analyzed after injecting recombinant angiostatin into the angeioma model
(cock wattle). Results Angiostatin showed high-efficiency expression in Pichia pastoris and 98% of recombinant an-
giostatin had been obtained after the purification. The recombinant angiostatin had activity on suppressing of angiogen-
esis on CAM and inducing of vascular endothelial cells apoptosis. It's 51% (P<0.01) of the inhibition rate of vascular
growth in the angeioma model and 39% (P<0.05) of the inhibition rate of proliferation of the hemangioma model after
14 days of maintenance therapy. Conclusion Drug of recombinant angiostatin had been successfully prepared. Angi-
ostatin possesses prominent effect on the treatment of angeioma.
