      【摘要】 目的 观察姜黄素对甲状腺癌细胞SW579侵袭力影响,并探讨姜黄素抗癌的作用机制。方法
状腺鳞癌SW579细胞株经5~20 μmol/L的不同浓度姜黄素作用48 h后,MTT法测定细胞增殖能力;流式细胞仪
检测细胞周期;RT-PCR方法检测细胞CD147及p21 mRNA的表达。结果 伴随姜黄素作用浓度的升高,SW579细
胞的生长呈显著抑制状态,呈剂量、时间依赖性。肿瘤细胞生长滞留在G1期,CD147 mRNA表达水平均显著下调,
而p21 mRNA表达水平均显著上调,呈剂量、时间依赖性。结论 姜黄素能有效抑制甲状腺癌细胞的增殖,并且
可能通过下调CD147的表达抑制肿瘤细胞的迁移。

Influence of curcumin on the proliferation and migration of thyroid carcinoma cells.

【Abstract】 Objective To observe the effect of curcumin on the invasion of human thyroid carcinoma cell SW579,
and to explore the mechanism of curcumin against cancer. Methods Thyroid squamous cell carcinoma cell line SW579
were cultured with different concentrations of curcumin 5~20 μmol/L for 48 h, and then to observe the change of cell growth
and morphology under inverted microscope; the proliferation was determined by MTT assay; cell cycle was detected by flow
cytometry; the expression of RT- PCR method for detection of CD147 and p21 in mRNA cells. Results Along with the in-
crease of the concentration of curcumin, the growth of SW579 cells was significantly inhibited, cell growth in G1, CD147 and
p21 mRNA expression levels were significantly increased, in a dose dependent manner. Conclusion Curcumin can effec-
tively inhibit the proliferation and migration of thyroid carcinoma cells.
