      【摘要】 目的 研究哮喘小鼠外周血CD8+CD28-T细胞与CD8+CD56+T细胞间的关系及两种细胞群在哮喘中的
作用。方法 30只BALB/c小鼠随机分为对照组和哮喘组,每组各15只。测定小鼠的气道反应性,对支气管肺
泡灌洗液行细胞学分类,观察肺组织的病理变化,流式检测小鼠外周血CD8+CD28-T细胞及CD8+CD56+T细胞的比
例,分析哮喘中两种细胞之间的相关性。结果 成功建立小鼠哮喘模型。①哮喘组小鼠BALF中细胞总数和嗜
酸性粒细胞(%)较对照组明显增高;②哮喘组小鼠外周血CD8+CD28-T细胞及CD8+CD56+T细胞比例均较对照组升
高;③ CD8+CD28-T细胞与CD8+CD56+T细胞数量上呈正相关(r=0.737,P=0.002)。结论 哮喘时外周血CD8+CD56+细
胞和CD8+CD28-细胞数目异常,两种细胞正相关,可能在哮喘发病中起重要作用。

Function and mechanism of CD8 +CD28- T cells and CD8 +CD28- T cells in peripheral blood of asthmatic mouse
model.

【Abstract】 Objective To examine the relationship between CD8 +CD28- and CD8 +CD56 + T cells in peripheral
blood of mice and the function of the two subsets in the development of asthma. Methods Thirty Balb/c mice were
randomly divided into two groups: asthma group, control group, with 15 mice in each group. The airway responsive-
ness were measured, bronchoalveolar lavage cytology was performed and lung tissue sections were prepared for histo-
pathologic examination to evaluate the airway inflammation, examine the fraction of CD8+CD28- and CD8+CD56+ T cells
by flow cytometry and correlation analysis. Results The mouse models established successfully. ① The total cell
number and the percentages of eosinophils in BALF of the asthmatic group were significantly higher than that in the
control group; ② The percentages of CD8 +CD28- and CD8 +CD56 + T cells in peripheral blood [(13.65±3.075) vs (4.11±
1.47)] in asthmatic group were significantly higher than that in control group [(10.32±2.318) vs (2.59±1.19)].③ There
was a strong positive correlation between CD8+CD28- and CD8+CD56+ T cells (r=0.737, P=0.002). Conclusion The frac-
tions of CD8+CD28- and CD8+CD56+ T cells are associated with asthma ongoing closely, and there is a strong positive cor-
relation between these two subsets.
