      【摘要】 目的 探讨金纳多 (Ginaton)对大鼠急性脊髓损伤 (ASCI)后脊髓组织内质网应激相关蛋白
GADD153/CHOP表达的影响。方法 选取健康SD大鼠64只,随机分为治疗组和对照组各 32只。所有大鼠均
以改良Allen's法制备脊髓损伤模型,治疗组在模型制备的基础上给予金纳多注射治疗,对照组给予同等剂量的
生理盐水。分别于致伤后3 h、1 d、3 d、7 d时各取8只,以改良Tarlov评分法评价其神经功能;评分后处死,取受损
节段脊髓行HE染色及免疫组化染色,观察形态学改变和CHOP表达的特点;并应用TUNEL方法检测神经细胞
凋亡情况。其量的评定借助于麦克奥迪数码医学图像分析系统A (Motic .med 6.0)软件,结果分别用累积光密度
(IOD)及细胞凋亡指数(AI)来表示。结果 所有实验大鼠脊髓组织均有不同程度出血、水肿、细胞凋亡及坏死。
治疗组Tarlov评分除3 h时间点外,其他各时间点Tarlov评分明显高于对照组,且差异有统计学意义(P<0.05)。治
疗组与对照组不同时间点CHOP表达、细胞凋亡,除3 h时间点外,其他各个时间点治疗组CHOP表达及细胞凋亡
均明显低于对照组,且差异有统计学意义(P<0.05)。结论 金纳多对急性脊髓损伤有保护作用,能改善损伤脊髓
的神经功能,这可能与其能够抑制CHOP表达及脊髓细胞凋亡有关。

Effect of Ginaton on expression of GADD153/CHOP in acute spinal cord injury rat model.

【Abstract】 Objective To explore the effect of Ginaton on the expression of endoplasmic reticulum stress
protein GADD153/CHOP in acute spinal cord injury rat model. Methods Sixty-four rats were randomly divided into
the treatment group and the control group, with 32 rats in each group. All rats were modeled by modified Allen's legal
system, and those of treatment group were given Ginaton injection on the basis of the modeling, while those of control
group were given equal dose of normal saline. Eight rats of each group were selected and reached respectively 3 h, 1 d,
3 d, 7 d after injury, improved Tarlov score was used to evaluate the nerve function. Then the rats were killed, and the
damaged segmental spinal were taken off and observed by HE, the IOD of CHOP were evaluated by immunohisto-
chemical method, the AI of cells were also evaluated by TUNEL method. Results There was different degree of hem-
orrhage, edema, apoptosis and necrosis on the spinal cord tissue of each group. The Tarlov scores of the treatment
group were all significantly higher than those of the control group in addition to 3 h after the injury (P<0.05), while
the CHOP IOD and AI of the treatment group were all significantly higher than those of the control group in addition
to 3 h after the injury (P<0.05). Conclusion Ginaton can improve the neural function and has protective effect on
acute spinal cord injury, which may related to its inhibition on CHOP expression and apoptosis of spinal cord.
