      标题:曲美他嗪对糖尿病大鼠心肌损伤的保护作用及机制探讨

      【摘要】 目的 探讨Caveolin-3(Cln-3)对1型糖尿病大鼠心肌损伤的影响。方法 26只雄性SD大鼠随机分
为对照(NC)组、糖尿病(DM)组、糖尿病+曲美他嗪 (Trimetazidine,TMZ)(DM+TMZ)、糖尿病+异丙肾上腺素
(Isoproterenol,ISO)(DM+ISO)组、糖尿病+异丙肾上腺素+曲美他嗪(DM+ISO+TMZ)组。糖尿病组65 mg/kg链脲
佐菌素(Streptozotocin,STZ)腹腔注射诱发1型糖尿病,异丙肾上腺素组在成模第4天进行4 mg/kg ISO腹腔注射
以加重心肌损伤,曲美他嗪组TMZ 40 mg/(kg·d)7 d灌胃观察其对1型糖尿病心肌损伤的疗效。比较各组大鼠血
清、心肌组织生化指标的变化,采用Western blot检测心肌Cln-3蛋白总含量。结果 NC组、DM组、DM+TMZ组
各项指标差异无统计学意义;DM+ISO组、DM+ISO+TMZ组与DM组比较,LDH、CK、MDA差异有显著统计学意
义(P<0.01),SOD差异有统计学意义(P<0.01~0.05),与NC组相比,DM组和DM+TMZ组心肌组织Cln-3蛋白表达差
异无统计学意义;与DM组比较,DM +ISO组、DM+ISO+TMZ组心肌组织Cln-3蛋白表达减少,差异有统计学意义
P<0.05),DM+ISO组与DM+ISO+TMZ组差异无统计学意义。结论 曲美他嗪可以保护心肌损伤的糖尿病大鼠。

br>Mechanism of protective effect of trimetazidine on myocardial injury in diabetic rats.

【Abstract】 Objective To study the effect of caveolin-3 on myocardial damage of Type 1 diabetic rats.
Methods 26 SD rats were randomly divided into five groups. These rats were provided streptozotocin (65 mg/kg) by
intraperitoneal injection to induce type 1 diabetes. When we got the diabetic rats, ISO(4 mg/kg) was provided through
intraperitoneal injection at the fourth day. TMZ (40 mg/kg) was provided through intragastric administration. After 1
week, the CK activities of serum and LDH, MDA and SOD contents were measured, the total protein content of caveo-
lin-3 was analyzed by western blot. Results Compared with the NC group, all the parameters had no significant dif-
ference in DM group, DM+TMZ group. Compared with the DM group, CK, LDH, MDA contents were significantly
increased (P<0.01), and SOD activities were significantly decreased(P<0.01, P<0.05)in DM+ISO group and DM+
ISO+TMZ group; compared between DM+ISO group and DM+ISO+TMZ group, CK, LDH, MDA contents were sig-
nificantly decreased (P<0.01), and SOD activities were significantly increased(P<0.05)in DM + ISO + TMZ group.
Compared with the DM group, the expression levels of caveolin-3 were significantly decreased(P<0.05)in DM+ISO
group and DM+ISO+TMZ group, but there were no significant difference between DM+ISO group and DM+ISO+
TMZ group. Conclusion TMZ can protect the myocardial damage in diabetic rats.
