      【摘要】 目的 探讨BRCA1相关蛋白 1 (BAP1)在肝细胞肝癌(LIHC)中的表达变化及其与预后的关系。方法 从UALCA获得癌症基因组图谱(TCGA)中LIHC的BAP1 mRNA表达水平及临床数据并进行数据分组和处理。采用R3.2.2软件进行分析。比较BAP1在癌组织与正常组织中的表达差异,并分析LIHC患者各亚组临床指标的BAP1表达水平与正常组织的差异。采用寿命表法计算生存率,采用Kaplan-Meier法比较BAP1高表达组和中低表达组患者的生存率,并绘制患者的生存曲线。结果 LIHC组织中BAP1 mRNA表达中位数为37.748 TPM,明显高于正常组织中的18.444 TPM,差异有显著统计学意义(P<0.01);患者的性别、年龄、种族、体质量、组织学类型、组织学分级、淋巴结、TNM分期Ⅰ~Ⅲ期、TP53突变的各组癌组织中BAP1表达水平与正常组织表达水平比较差异均有统计学意义(P<0.05),而Ⅳ期组癌组织中BAP1表达水平与正常组织表达水平比较差异无统计学意义(P>0.05);患者的中位生存时间为27.18个月,1年、2年、3年、4年、5年生存率分别为0.57%、0.31%、0.20%、0.13%、0.08%;BAP1 mRNA高表达的整体生存率较BAP1mRNA中低表达者短,差异均有统计学意义(P<0.05)。结论 BAP1 mRNA在LIHC中呈高表达,高表达组生存率较低,提示预后不良。

Expression of BAP1 in liver hepatocellular carcinoma and its relationship with prognosis.

【Abstract】 Objective To analyze the expression of BRCA1 associated protein 1 (BAP1) in liver hepatocellularcarcinoma (LIHC) and its relationship with prognosis.Methods BAP1 mRNA expression levels and clinical data of LI-HC in The Cancer Genome Atlas (TCGA) were obtained from UALCA, and the data were grouped and processed.R3.2.2 software was used for analysis. The expression difference of BAP1 in tumor tissue and normal tissue was com-pared, and the difference of BAP1 expression in clinical data was analyzed. Life table method was used to calculate thesurvival rate. Kaplan-Meier method was used to compare the survival rate of patients with high and low BAP1 expres-sion group, and the survival curve of patients was drawn. Results The median expression of BAP1 mRNA in LIHC tis-sues was 37.748 TPM, which was significantly higher than 18.444 TPM in normal tissues (P<0.01). The expression levelof BAP1 in tumor tissues of patients with different gender, age, race, body mass, histological type, histological grade,lymph nodes, TNM stage Ⅰ to Ⅲ, and TP53 mutation was significantly different from that in normal tissues (P<0.05).There was no significant difference in the expression level of BAP1 between stage Ⅳ tumor tissues and normal tissues(P>0.05). The median survival time was 27.18 months, and the 1-year, 2-year, 3-year, and 4-year survival rates wererespectively 0.57%, 0.31%, 0.20%, 0.13%, and 0.08%. The overall survival rate of patients with high expression ofBAP1 mRNA was significantly lower than that in patients with medium and low expression of BAP1 mRNA (P<0.05).Conclusion BAP1 mRNA is highly expressed in LIHC, and the survival rate of patients with high expression of BAP1mRNA is lower, suggesting poor prognosis.
