      标题:黄芪多糖对大鼠急性胰腺炎的作用及其机制研究

      【摘要】 目的 探索黄芪多糖对大鼠急性胰腺炎的作用及其潜在机制,以期为急性胰腺炎的治疗提供新思路。方法 将60只大鼠随机分为对照组、模型组、治疗组,每组20只。将牛磺胆酸钠注射入大鼠胰胆管的方法构建重症急性胰腺炎大鼠模型,治疗组大鼠造模后灌胃给药400 mg/(kg·d)的黄芪多糖,对照组和模型组大鼠灌胃给药等量生理盐水,2周后处死大鼠收集胰腺和血清。采用H&E染色分析黄芪多糖对不同实验组大鼠胰腺组织结构的影响,采用酶联免疫吸附法检测血清中白介素-6 (IL-6)、白介素1β (IL-1β)和肿瘤坏死因子α (TNF-α)表达水平;采用蛋白免疫印迹实验检测 p-NF-κB p65、NF-κB p65、IKKα/β、p-IKKα/β和 IKB-α表达水平。结果 与模型组比较,黄芪多糖能够减少急性胰腺炎大鼠胰腺组织坏死灶和炎症细胞浸润区域,减轻急性胰腺炎大鼠胰腺组织的组织学病变;黄芪多糖治疗组大鼠的 TNF-α,IL-6和 IL-1β水平分别为(27.18±0.33) pg/mL、(92.17±1.33) pg/mL、(150.68±5.75) pg/mL,明显低于模型组的(47.92±1.29) pg/mL、(152.92±2.59) pg/mL、(239.88±3.12) pg/mL,差异均有统计学意义(P<0.05);黄芪多糖治疗组大鼠的NF-κB p65和 IKKα/βJAK2蛋白磷酸化水平分别为0.31±0.02、0.29±0.03,明显低于模型组的1.58±0.05、1.31±0.09,差异均有统计学意义(P<0.05);黄芪多糖治疗组大鼠的IKB-α的表达水平为0.78±0.01,明显高于模型组的0.23±0.0,差异均有统计学意义(P<0.05)。结论 黄芪多糖可减轻急性胰腺炎大鼠炎性反应,且可能与抑制NF-κB3通路相关。

Effect of Astragalus polysaccharides on acute pancreatitis in rats and its mechanism.

【Abstract】 Objective To explore the effect of Astragalus polysaccharides (APS) on acute pancreatitis in ratsand its potential mechanism, in order to provide new ideas for the treatment of acute pancreatitis. Methods Sixty ratswere randomly divided into a control group, a model group, and a treatment group, with 20 rats in each group. Rat mod-els of severe acute pancreatitis were established by injecting sodium taurocholate into the pancreatic bile duct of rats.The rats in the treatment group were given 400 mg/(kg·d) of Astragalus polysaccharides by gavage after modeling,while the rats in the control group and model group were given an equal amount of physiological saline by gavage. After2 weeks, the rats were sacrificed to collect pancreatic glands and serum. H&E staining was used to analyze the effect ofAstragalus polysaccharides on the pancreatic tissue structure of rats in different experimental groups, and enzyme-linkedimmunosorbent assay was used to detect the levels of interleukin-6 (IL-6), interleukin-1β (IL-1β), and tumor necrosisfactor α (TNF-α) in serum. Western blot was used to detect the levels of p-NF-κB p65, NF-κB p65, IKKα/β, P-IKKα/β,and IKB-α. Results Compared with the model group, Astragalus polysaccharides can reduce the necrotic foci and in-flammatory cell infiltration areas of pancreatic tissue in rats with acute pancreatitis, and alleviate the histological changesof pancreatic tissue in rats with acute pancreatitis. TNF-α, IL-6, and IL-1β levels in the treatment group were (27.18±0.33) pg/mL, (92.17±1.33) pg/mL, and (150.68±5.75) pg/mL, which were significantly lower than (47.92±1.29) pg/mL,(152.92±2.59) pg/mL, and (239.88±3.12) pg/mL of the model group (P<0.05); phosphorylation levels of NF-κB p65 andIKKα/βJAK2 protein in the treatment group were 0.31±0.02 and 0.29±0.03, respectively, significantly lower than 1.58±0.05 and 1.31±0.09 in the model group (P<0.05); IKB-α level in the treatment group was 0.78±0.01, significantly higherthan 0.23±0.0 in the model group (P<0.05). Conclusion Astragalus polysaccharides can alleviate the inflammatory re-sponse in rats with acute pancreatitis and may be associated with the inhibition of NF-κB3 pathway.
