      【摘要】 GPR30是一种G蛋白偶联受体,同时其因为能够与雌激素直接结合而被称为G蛋白偶联雌激素受体。作为雌激素的新型受体,其功能被广泛与雌激素已知效应比较。雌激素对神经中枢系统作用以及这些作用是否能够转化用于治疗各种神经系统疾病已经得到广泛的讨论。据文献报道GPR30受体在神经中枢系统中分布并部分参与雌激素样神经保护作用,同时其被激动不具备雌激素直接使用带来的副作用,比如乳腺癌,雌化效应。这些特征提示GPR30受体可能是新的从激素角度出发治疗神经系统疾病的潜在靶点。本综述首先总结该受体在脑部分布,介导的信号通路以及这些通路产生的神经作用,随后对近十年关于GPR30神经作用应用于神经系统疾病时获取的新认识进行总结并从中提出针对神经系统疾病可能的治疗策略,最后粗略探讨GPR30受体在神经系统疾病临床转化过程中可能遇到的问题。

Role of GPR30 receptor in neurodegenerative diseases and cerebral ischemia.

The First Hospitalof Jiaxing, Jiaxing 314000, Hangzhou, CHINA【Abstract】 GPR30, a G protein-coupled receptor, is also known as a G protein-coupled estrogen receptor due toits direct binding with estrogens. As a novel receptor for estrogens, its functions have been extensively compared to theknown effects of estrogens. The effects of estrogens on the central nervous system and their potential therapeutic applica-tions for various neurological disorders have been widely discussed. Literature indicate that GPR30 receptors are distrib-uted in the central nervous system and partially involved in estrogen-like neuroprotective effects, without the side effectsassociated with direct estrogen usage, such as breast cancer and feminizing effects. These characteristics suggest thatGPR30 receptors may serve as potential targets for hormone-based therapies in neurological disorders. In this review, wefirst summarize the distribution of this receptor in the brain, the signaling pathways it mediates, and the neural effectsproduced by these pathways. Subsequently, we provide an overview of the recent insights gained from studying the neu-rological effects of GPR30 over the past decade and propose potential therapeutic strategies for neurological disorders.Finally, we briefly discuss the potential challenges associated with the clinical translation of GPR30 receptors in the fieldof neurological disorders.
