      【摘要】 骨质疏松(OP)是一种全身性骨骼疾病,其特点是骨量减少及骨组织微结构的退变。炎症和免疫调控破骨细胞和成骨细胞的功能在骨质疏松发生发展中起重要作用。NLRP3炎性小体是细胞内能够识别多种多样的内源性及外源性危险信号,并与接头蛋白(ASC)以及半胱氨酸蛋白酶-1 (Caspase-1)组装形成一种被称为“炎性小体”的多蛋白复合体,使促炎因子成熟(如 IL-1β和 IL-18),引起细胞焦亡等病理改变。现就NLRP3引起的细胞焦亡(pyroptosis)在骨质疏松中的作用及最新研究进展进行综述,以期为骨质疏松的治疗提供一种新的策略,为指导骨质疏松药物研发提供理论依据。

Research progress of NLRP3 inflammasome in osteoporosis.

Inflammation and immune regulation of osteoclasts and osteoblasts play an important role inthe development of osteoporosis. Nucleotide-binding domain and leucine-rich repeat protein-3 (NLRP3) inflammasomeis a multiprotein complex that can recognize a wide variety of endogenous and exogenous danger signals and assemblewith caspase recruitment domain (ASC) and caspase-1. It can help the pro-inflammatory factors mature (such as IL-1βand IL-18) and cause pyroptosis and other pathological. This article summarizes the role of NLRP3-induced pyroptosisin osteoporosis and the latest research progress, so as to provide a new strategy for the treatment of osteoporosis and pro-vide theoretical basis for the development of osteoporosis drugs.
