      标题:右美托咪定调控circ_0072995对直肠癌细胞SW480增殖凋亡的研究

      【摘要】 目的 探讨右美托咪定对直肠癌细胞增殖、凋亡的影响及分子机制。方法 收集2017年1月至2021年1月宝鸡市人民医院收治的88例直肠癌患者的癌组织及癌旁组织标本。采用实时荧光定量PCR (RT-qPCR)检测肠癌组织和细胞中 circ_0072995的表达水平;分析 circ_0072995表达与临床病理关系。SW480细胞分为con组、Dex1 μmol/L组、Dex 2 μmol/L组、Dex 5 μmol/L组、si-circ_0072995组、si-NC组、Dex+pcDNA-circ_0072995组、Dex+pcDNA组;MTT法检测细胞增殖抑制率;克隆形成实验检测克隆形成数;流式细胞术检测细胞凋亡率。结果 直肠癌组织中circ_0072995表达水平为2.36±0.15,明显高于癌旁组织的1.00±0.02,差异有统计学意义(P<0.05);circ_0072995表达水平与TNM分期及淋巴结转移有关(P<0.05);不同浓度右美托咪定处理后SW480细胞增殖抑制率和凋亡率升高,克隆形成数减少,circ_0072995表达水平降低,呈浓度依赖性,差异均有统计学意义(P<0.05);沉默 circ_0072995后,SW480细胞增殖抑制率和凋亡率升高,克隆形成数减少,差异均有统计学意义(P<0.05);与Dex+pcDNA组比较,Dex+pcDNA-circ_0072995组 circ_0072995表达水平升高,细胞增殖抑制率和凋亡率降低,克隆形成数增加,差异均有统计学意义(P<0.05)。结论 右美托咪定可通过下调 circ_0072995抑制直肠癌细胞增殖,促进凋亡。

Dexmedetomidine regulates the proliferation and apoptosis of rectal cancer cell SW480 by circ_0072995.

【Abstract】 Objective To explore the effect of dexmedetomidine on the proliferation and apoptosis of rectalcancer cells and its molecular mechanism. Methods The cancer tissues and paracancerous tissues of 88 patients withcolorectal cancer from January 2017 to January 2021 in Baoji People's Hospital were collected. Real-time fluorescentquantitative PCR (RT-qPCR) was used to detect the expression level of circ_0072995 in colorectal cancer tissues andcells. The relationship between circ_0072995 expression and clinicopathology was analyzed. SW480 cells were dividedinto con group, Dex 1 μmol/L group, Dex 2 μmol/L group, Dex 5 μmol/L group, si-circ_0072995 group, si-NC group,Dex+pcDNA-circ_0072995 group, Dex+pcDNA group. MTT method was used to detect cell proliferation inhibitionrate. Clone formation test was used to detect the number of clones formed. Flow cytometry was used to detect the apopto-sis rate. Results The expression level of circ_0072995 in rectal cancer tissues was higher than that in adjacent tissues:(2.36±0.15) vs (1.00±0.02), P<0.05. The expression level of circ_0072995 was correlated with TNM stage and lymphnode metastasis (P<0.05). After treatment with different concentrations of dexmedetomidine, the proliferation inhibi-tion rate and apoptosis rate of SW480 cells increased, the number of clones decreased, and the expression level ofcirc_0072995 was decreased, in a concentration-dependent manner, with statistically significant differences (P<0.05). Af-ter silencing circ_0072995, the proliferation inhibition rate and apoptosis rate of SW480 cells were increased, and thenumber of clones was decreased, with statistically significant differences (P<0.05). Compared with the Dex+pcDNAgroup, the expression level of circ_0072995 in the Dex+pcDNA-circ_0072995 group was increased, the cell prolifera-tion inhibition rate and apoptosis rate were decreased, and the number of clones was increased, with statistically signifi-cant differences (P<0.05). Conclusion Dexmedetomidine can inhibit the proliferation of rectal cancer cells and pro-mote apoptosis by down-regulating circ_0072995.
