      标题:多囊卵巢综合征患者颗粒细胞中GAS5、miR-222的表达及其相关性

      【摘要】 目的 研究长链非编码RNA GAS5 (lncRNA GAS5)和微小RNA-222 (miR-222)在多囊卵巢综合征(PCOS)患者颗粒细胞中的表达情况,初步探讨PCOS患者GAS5与miR-222的相关性。方法 选取2019年12月至2020年 10月于深圳市妇幼保健院生殖医学中心接受体外受精-胚胎移植(IVF-ET)治疗的 86例不孕症患者,其中42例典型的PCOS患者纳入实验组,其余44例因男方因素不孕行 IVF-ET患者纳入对照组,应用实时荧光定量聚合酶链式反应(qRT-PCR)检测两组受试者颗粒细胞中GAS5和miR-222的表达情况。比较两组患者的一般资料及性激素六项水平,采用Pearson相关分析法分析PCOS患者GAS5与性激素和miR-222的相关性。结果 实验组患者身体质量指数(BMI)、黄体生成素(LH)及孕酮(P)明显高于对照组,差异均有统计学意义(P<0.05);与对照组比较,实验组患者颗粒细胞中的GAS5表达水平明显降低,miR-222表达水平明显升高,差异均有统计学意义(P<0.05);经 Pearson相关性分析结果显示,GSA5与miR-222表达呈负相关(r=-0.545,P=0.000 2),GAS5与 LH呈负相关(r=-0.89,P=0.019)。结论 PCOS患者颗粒细胞中GAS5及miR-222表达异常,GAS5与miR-222表达和LH呈负相关,预示GAS5和miR-222可能在PCOS的发病机制中发挥重要作用。

Expression and correlation of long-chain non-coding RNA GAS5 and miR-222 in granulosa cells of patientswith polycystic ovary syndrome.

【Abstract】 Objective To study the expression of long-chain non-coding RNA GAS5 (lncRNA GAS5) and mi-croRNA-222 (miR-222) in granulosa cells of patients with polycystic ovary syndrome (PCOS), and to preliminarily ex-plore the correlation between GAS5 and miR-222 in patients with PCOS. Methods A total of 86 patients with infertili-ty who underwent in vitro fertilization-embryo transfer (IVF-ET) treatment in the Reproductive Medicine Center, Shen-zhen Maternity and Child Health Hospital Affiliated to Southern Medical University from December 2019 to October2020 were selected. Among them, 42 typical PCOS patients were included in the experimental group, and the remaining44 patients who underwent IVF-ET due to male infertility were included in the control group. Real-time fluorescent quanti-tative polymerase chain reaction (qRT-PCR) was used to detect the expression of GAS5 and miR-222 in the granulosacells between the two groups of subjects. The general data and levels of six sex hormone were compared between the twogroups. Pearson correlation analysis method was used to analyze the correlation between GAS5 and sex hormones,miR-222 in PCOS patients. Results The body mass index (BMI), luteinizing hormone (LH), and progesterone (P) in theexperimental group were significantly higher than those in the control group, and the differences were statistically signifi-cant (P<0.05). Compared with the control group, the expression level of GAS5 in granulosa cells in the experimentalgroup was significantly reduced, and the expression level of miR-222 was significantly increased, with statistically signifi-cant difference (P<0.05). Pearson correlation analysis showed that the expression of GSA5 and miR-222 was negativelycorrelated (r=-0.545, P=0.000 2), GAS5 was negatively correlated with LH (r=-0.89, P=0.019). Conclusion The expres-sion of GAS5 and miR-222 is abnormal in the granular cells of PCOS patients. GAS5 is negatively correlated with the ex-pression of miR-222 and LH, indicating that GAS5 and miR-222 may play an important role in the pathogenesis of PCOS.
