      标题:miR-196a及miR-1285在上皮性卵巢癌中的表达及其诊断价值研究

      【摘要】 目的 研究miR-196a及miR-1285在上皮性卵巢癌中的表达及其诊断价值。方法 选取安康市妇幼保健院于2019年7月至2021年4月收治的95例拟行手术治疗的上皮性卵巢癌患者作为研究组,选取同期本院收治的上皮性卵巢良性肿瘤患者110例作为良性组,选取同期在本院体检的正常健康女性110例作为对照组。采用RT-PCR法测定各组受检者血浆中miR-196a及miR-1285表达水平。分析miR-196a及miR-1285表达水平与上皮性卵巢癌临床病理特征的关系,采用受检者工作特征曲线(ROC)分析miR-196a及miR-1285表达水平对上皮性卵巢癌的诊断价值,采用多元Logistic回归模型分析miR-196a及miR-1285表达水平与上皮性卵巢癌的关系,采用Pearson相关分析法分析上皮性卵巢癌患者血浆中miR-196a表达水平与miR-1285表达水平的相关性。结果 对照组、良性组和研究组受检者血浆miR-196a及miR-1285表达水平组间比较差异均具有统计学意义(P<0.05);研究组患者血浆miR-196a表达水平明显高于对照组和良性组,血浆miR-1285表达水平明显低于对照组和良性组,差异均有统计学意义(P<0.05);对照组和良性组患者血浆miR-196a和miR-1285表达水平组间比较差异均无统计学意义(P>0.05);上皮性卵巢癌患者的血浆miR-196a及miR-1285表达水平与病理分期均有关(P<0.05),而与患者年龄、病理类型、病理分级、术后是否有肿瘤残余等因素均无关(P>0.05);上皮性卵巢癌患者血浆miR-196a的曲线下面积(AUC)及95%CI分别为0.839 (0.785~0.894,P<0.001),miR-1285的AUC及95%CI分别为0.858 (0.810~0.905,P<0.001);多元Logistic回归模型分析结果显示,血浆miR-196a表达水平升高是上皮性卵巢癌的独立危险因素(P<0.05),miR-1285表达水平降低是上皮性卵巢癌的独立保护因素(P<0.05);经Pearson相关性分析结果显示,血浆miR-196a与miR-1285呈负相关(r=-0.658,P<0.05)。结论 上皮性卵巢癌患者血浆中miR-196a表达水平升高,miR-1285表达水平降低,两者对上皮性卵巢癌均具有一定的诊断价值。

【Abstract】 Objective To study the expression and diagnostic value of miR-196a and miR-1285 in epithelialovarian cancer. Methods Ninety-five patients with epithelial ovarian cancer prepared to be treated by surgery in Ank-ang Maternal and Child Health Hospital from July 2019 to April 2021 were selected as the study group, 110 patients withbenign epithelial ovarian tumors treated in the same period were selected as the benign group, and 110 normal healthywomen during the same period were selected as the control group. RT-PCR was used to determine the expression levelsof miR-196a and miR-1285 in the plasma of each group of subjects. The relationship between the expression levels ofmiR-196a and miR-1285 and the clinicopathological features of epithelial ovarian cancer were analyzed. The diagnosticvalue of miR-196a and miR-1285 expression levels in epithelial ovarian cancer was analyzed by receiver operating char-acteristic (ROC) curve. Multiple logistic regression model was used to analyze the relationship between the expressionlevels of miR-196a and miR-1285 and epithelial ovarian cancer. Pearson correlation analysis was used to analyze thecorrelation between the expression level of miR-196a and miR-1285 in the plasma of patients with epithelial ovariancancer. Results There were statistically significant differences in plasma expression levels of miR-196a and miR-1285among the control group, benign group and study group (P<0.05).
