      【摘要】 目的 探究miR-147a在膀胱癌组织和细胞中的表达及对膀胱癌细胞增殖及细胞侵袭能力的影响。方法 通过实时荧光定量PCR (qRT PCR)法检测空军军医大学第二附属医院检验科2020年1~12月期间采集到的30例膀胱癌组织细胞与正常组织细胞中miR-147a表达水平,分别将miR-147a mimics序列和miR NC序列转染至人膀胱移行细胞癌细胞系 um-uc-3,作为miR-147a mimics组和miR NC组,通过Westernblot实验检测转染效率,采用MMT实验和Transwell实验检测并比较两组um-uc-3细胞增殖、迁移以及侵袭能力。结果 膀胱癌组织和细胞的miR-147a表达水平分别为(0.37±0.06)%、(0.41±0.08)%,明显低于正常膀胱组织的(1.20±0.15)%和细胞的(1.69±0.25)%,差异均有统计学意义(P<0.05);miR-147a mimics组转染后 72 h、96 h的OD值分别为(0.75±0.26)%、(1.30±0.38)%,明显低于miR-NC组的(1.06±0.34)%、(1.71±0.46)%,差异均有统计学意义(P<0.05);miR-147a mimics组的细胞迁移数、细胞侵袭数分别为(123.06±25.16)%、(91.48±16.74)%,明显低于miR-NC组的(314.75±68.52)%、(252.30±57.23)%,差异均有统计学意义(P<0.05)。结论 在膀胱癌组织和细胞中,miR-147a的表达均有不同程度的下调,体外上调miR-147a表达可有效抑制膀胱癌细胞的增殖、迁移和侵袭能力。

Expression of miR-147a in bladder cancer tissues and cells and its effect on proliferation and migration ofbladder cancer cells.

【Abstract】 Objective To investigate the expression of miR-147a in bladder cancer tissues and cells and its ef-fect on proliferation and invasion of bladder cancer cells. Methods Real-time fluorescent quantitative PCR (qRT PCR)was used to detect the expression level of miR-147a in 30 cases of bladder cancer tissues and normal tissues collected bythe Department of Laboratory at the Second Affiliated Hospital of Air Force Medical University from January to Decem-ber 2020. The miR-147a mimics sequence and miR-NC sequence were transfected into the human bladder transitional cellcarcinoma cell line um-uc-3, respectively, as the miR-147a mimics group and the miR-NC group. The transfection effi-ciency was detected by Western blot assay; the proliferation, migration and invasion abilities of um-uc-3 cells were com-pared in the two groups by MMT assay and Transwell assay. Results The expression levels of miR-147a in bladder can-cer tissues and cells were (0.37±0.06)% and (0.41±0.08)%, respectively, which were significantly lower than that in nor-mal bladder tissues of (1.20±0.15)% and cells of (1.69±0.25)% (P<0.05). The OD values at 72 h and 96 h after transfec-tion were (0.75±0.26)% and (1.30±0.38)% in miR-147a mimics group, respectively, which were significantly lower thancorresponding (1.06±0.34)% and (1.71±0.46)% in the miR-NC group (P<0.05). The number of cell migration and invasionwere (123.06±25.16)% and (91.48±16.74)% in the miR-147a mimics group, respectively, which were lower than (314.75±68.52)% and (252.30±57.23)% in the miR-NC group, with statistically significant differences (P<0.05). Conclusion Inbladder cancer tissues and cells, the expression of miR-147a is down-regulated to varying degrees. Up-regulation ofmiR-147a expression in vitro can effectively inhibit the proliferation, migration and invasion of bladder cancer cells.
