      标题:喉鳞状细胞癌组织中 IDO1和PD-L1的表达及其与临床病理特征及预后的关系

      【摘要】 目的 检测吲哚胺2,3-双加氧酶1 (IDO1)和程序性死亡配体-1 (PD-L1)在喉鳞状细胞癌(LSCC)患者肿瘤组织中的表达情况,探讨其与LSCC病理特征及预后的关系。方法 收集2012年6月至2013年6月期间榆林市第二医院收治的LSCC患者的癌组织标本85例以及对应癌旁的正常组织标本28例。通过免疫组化法检测标本中 IDO1和PD-L1表达水平,单因素分析两者阳性表达率与LSCC患者的临床病理参数的关系,Pearson相关性分析两者阳性表达的相关性。Kaplan-Meier法分析 IDO1和PD-L1表达与患者预后的关系。结果 IDO1和PD-L1在LSCC组织中阳性表达率分别为62.35%和55.29%,明显高于癌旁正常组织中的35.71%和21.43%,差异均有统计学意义(P<0.05);单因素分析结果显示,LSCC组织中 IDO1和PD-L1的阳性表达率与肿瘤分化程度、TNM分期、淋巴结转移有关(P<0.05);Pearson相关性分析结果显示,LSCC组织中 IDO1与PD-L1的阳性表达呈正相关(r=0.527,P=0.021);Kaplan-Meier法分析发现 IDO1和PD-L1低表达组患者的平均总生存时间分别为45.5个月和46.3个月,长于高表达组的39.6个月和38.8个月,差异有统计学意义(P<0.05)。结论 IDO1和PD-L1在LSCC组织中的阳性表达率显著增加,且与患者临床病理特征及预后有关,有可能作为LSCC患者诊疗靶标和预后标志物。

Expression of IDO1 and PD-L1 in laryngeal squamous cell carcinoma and their relationship withclinicopathological features and prognosis.

【Abstract】 Objective To detect the expression of indoleamine 2, 3-dioxygenase 1 (IDO1) and programmeddeath ligand-1 (PD-L1) in laryngeal squamous cell carcinoma (LSCC) patients with tumor tissue, and to investigate therelationship between LSCC and its pathological characteristics and prognosis. Methods A total of 85 cancer tissuespecimens of LSCC patients and 28 corresponding normal tissue samples were collected from June 2012 to June 2013 inYulin Second Hospital. The expression of IDO1 and PD-L1 proteins in LSCC was detected by immunohistochemistry.Univariate analysis was used to analyze the relationship between the positive expression rates of both and clinicopatho-logical parameters of LSCC patients. Pearson correlation analysis was used to explore the correlation between the twopositive expressions. Kaplan-Meier method was used to analyze the relationship between the expression of IDO1 andPD-L1 and the prognosis of patients. Results The positive expression rates of IDO1 and PD-L1 in LSCC tissues were62.35% and 55.29%, respectively, which were significantly higher than 35.71% and 21.43% in normal parcancerous tis-sues (P<0.05). Univariate analysis showed that the positive expression rates of IDO1 and PD-L1 in LSCC tissues were cor-related with tumor differentiation, TNM stage, and lymph node metastasis (P<0.05). Pearson correlation analysis showedthat there was positive correlation between the positive expression of IDO1 and PD-L1 in LSCC tissues (r=0.527, P=0.021). Kaplan-Meier analysis showed that the mean overall survival time of LSCC patients in IDO1 and PD-L1 low ex-pression groups were 45.5 months and 46.3 months, respectively, which were significantly longer than 39.6 months and38.8 months in high expression groups (P<0.05). Conclusion The positive expression rates of IDO1 and PD-L1 inLSCC tissues were significantly increased, which were related to the clinicopathological characteristics and prognosis ofLSCC patients. It may be used as the diagnosis and treatment targets and prognostic markers of LSCC patients.
