      标题:低剂量阿托伐他汀钙对线虫寿命及相关指标的影响

      【摘要】 目的 研究阿托伐他汀钙对秀丽隐杆线虫(简称“线虫”)寿命及相关指标的影响,了解其抗衰老作用。方法 使用野生型N2秀丽隐杆线虫株进行实验。将线虫分为空白对照组(阿托伐他汀钙浓度为0 μmol/L)和药物组(阿托伐他汀钙浓度分别为0.1 μmol/L、0.5 μmol/L、1 μmol/L及10 μmol/L)。通过普通寿命实验、铜绿假单胞菌寿命实验、热应激实验、咽泵和身体摆动率的测量、生殖实验以及生殖应激实验来评价阿托伐他汀钙对线虫寿命及相关指标的影响。结果 ①普通寿命实验:各药物组与对照组的平均寿命比较差异均无统计学意义(P>0.05);②铜绿假单胞菌寿命实验:与对照组的(10.00±2.05) d相比,0.5 μmol/L、1 μmol/L和10 μmol/L组平均寿命分别延长5.9%、6.06%和9.15%,差异均具有统计学意义(P<0.05);③热应激寿命实验:与对照组的(4.49±22.40) h相比,0.5 μmol/L、1 μmol/L和10 μmol/L组平均寿命分别延长21.16%、32.52%和50.78%,差异均有统计学意义(P<0.05);④咽泵频率及身体摆动频率测量:0.5 μmol/L和1 μmol/L组的咽泵频率和身体摆动频率明显高于对照组,差异均有统计学意义(P<0.05);⑤生殖实验:各药物组的线虫平均产卵量分别与对照组比较差异均无统计学意义(P>0.05);⑥热应激生殖实验:与对照组的(119.69±23.93)枚相比,1 μmol/L、10 μmol/L组分别为(139.07±17.01)枚和(141.42±24.69)枚,差异均有统计学意义(P<0.05)。结论 较低浓度的阿托伐他汀钙虽可提高线虫对抗应激的能力和生存质量,但并未影响线虫的寿命和生殖;既不促进也不延缓机体衰老,也不损害其生殖能力。

Effects of low dose atorvastatin calcium on the lifespan and its related indicators of C.

【Abstract】 Objective To study the effect of atorvastatin calcium on lifespan and related indexes of Caenorhab-ditis elegans (C. elegans) and to understand its anti-aging effect. Methods Wild type N2 C. elegans strains were used inthe experiment. The strains were divided into blank control group (the concentration of statins was 0 μmol/L) and druggroup (the concentrations of statins were 0.1 μmol/L, 0.5 μmol/L, 1 μmol/L, and 10 μmol/L, respectively). Ordinary lifetest, life test of Pseudomonas aeruginosa, heat stress life test, pharynx-pumping and body-swing rate measurement, repro-ductive test, and reproductive stress test were used to evaluate the effect of atorvastatin calcium on the life expectancyand the related indexes of nematode. Results ① Ordinary life test: the average life span of each drug group was not sig-nificantly different from that of the control group (P>0.05). ② Life test of pseudomonas aeruginosa: compared with thecontrol group (10.00±2.05) days, the average life span of 0.5 μmol/L, 1 μmol/L and 10 μmol/L group were prolonged by5.9%, 6.06%, and 9.15%, respectively, and the differences were statistically significant (P<0.05).③ Heat stress life test:compared with the control group (4.49 ± 22.40) hours, the average life span of 0.5 μmo/L, 1 μmol/L and 10 μmol/Lgroups were prolonged by 21.16%, 32.52%, and 50.78%, respectively, and the differences were statistically significant (P<0.05).④ Pharynx-pumping and body-swing rate measurement: the pharynx-pumping rate and body-swing rate of 0.5 μmol/Land 1 μmol/L group were higher than those of control group, and the differences were statistically significant (all P<0.05); ⑤ Reproduction test: compared with the control group, the drug groups showed no significant differences in theaverage number of eggs (P>0.05). ⑥ Reproductive stress test: the average number of eggs was (119.69±23.93) eggs inthe control group versus (139.07±17.01) eggs in 1 μmol/L group and (141.42±24.69) eggs in 10 μmol/L group, and thedifferences were statistically significant (P<0.05). Conclusion Low concentration of atorvastatin calcium can improvethe ability of the nematode resistant to stress and improve the quality of life of nematodes, but did not affect lifespan ofthe worm, reproduction, or senescence.
