      【摘要】 糖尿病肾病(DKD)是糖尿病的常见并发症,为终末期肾脏病的主要病因之一。DKD中糖代谢障碍所致炎症、氧化应激及糖基化终末产物等改变不同程度促使转化生长因子-β1 (TGF-β1)表达增加,加速病情进展。其中,TGF-β1与其受体结合并激活下游信号蛋白发挥生物学作用。Smads蛋白作为下游信号蛋白与TGF-β1组成了经典信号传导通路,参与了肾纤维化的病理过程。为进一步延缓肾纤维化进程,提升DKD治疗效果,必须全面了解TGF-β1/Smad信号通路在DKD的发病机理中涉及的分子机制。因此,本文概述了TGF-β1/Smad信号通路在DKD发病中的最新发现。

Research progress of relationship between TGF-β 1/Smad signaling pathway and diabetic nephropathy.

Department of Nephrology, the Fifth Afiliated (Zhuhai) Hospital of Zunyi Medical University,Zhuhai 519000, Guangdong, CHINA【Abstract】 Diabetic kidney disease (DKD) is a common complication of diabetes and one of the main causes ofend-stage kidney disease. The changes of hyperglycemia, oxidative stress and inflammation caused by carbohydrate me-tabolism disorders in DKD increase the expression levels of transforming growth factors (TGF) in varying degrees andaccelerate the progression of the disease. Among them, TGFs play a biological role by binding to its receptors and acti-vating downstream signaling proteins. The Smads protein, as a downstream signaling protein, constitutes a classical sig-naling pathway with TGF-β1, and participates in the pathological process of renal fibrosis. For purpose of further delay-ing the process of renal fibrosis and improving the therapeutic effect of DKD, it is necessary to fully understand the mo-lecular mechanism involved in the pathogenesis of the TGF-β 1/Smad signaling pathway in DKD. Therefore, this papersummarizes the latest findings of the TGF-β1/Smad signaling pathway in DKD pathogenesis.
