      标题:扶芳藤总三萜对衰老小鼠血清及组织T-AOC、CAT、GSH-PX表达的影响及意义

      【摘要】 目的 探索扶芳藤总三萜对D-半乳糖诱导的小鼠衰老模型抗氧化作用。方法 取健康清洁级雄性昆明小鼠75只,以皮下注射D-半乳糖(D-galactose)方法建立衰老小鼠模型;并将小鼠以单纯随机抽样法分为阳性药物组、模型组、扶芳藤总三萜高、低剂量组及正常对照组,每组15只,各组每天按预设方案给药连续21 d后,检测小鼠血清、心脏、肝脏及骨骼肌组织总抗氧化能力(T-AOC)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)三项抗氧化指标。结果 与对照组比较,高、低剂量组小鼠的血清 T-AOC及GSH-Px水平均升高,其中低剂量组的T-AOC水平明显高于对照组,差异有统计学意义(P<0.05),但其他各组的T-AOC及GSH-Px水平升高不明显,差异均无统计学意义(P>0.05);与对照组比较,高、低剂量组小鼠心脏组织的T-AOC、CAT、GSH-Px水平均升高,其中高、低剂量组的T-AOC水平明显高于对照组,高剂量组CAT水平明显高于对照组,差异均有统计学意义(P<0.05),但其他各组的T-AOC、CAT、GSH-Px水平升高不明显,差异均无统计学意义(P>0.05);与对照组比较,高、低剂量组小鼠肝脏组织的T-AOC、CAT、GSH-Px水平均升高,其中高剂量组的CAT水平明显高于对照组,高剂量组的GSH-Px水平明显高于对照组,差异均有统计学意义(P<0.05),但其他各组的T-AOC、CAT、GSH-Px水平升高不明显,差异均无统计学意义(P>0.05);与对照组比较,高、低剂量组小鼠骨骼肌组织的T-AOC、CAT、GSH-Px水平均升高,其中高、低剂量组的T-AOC水平明显高于对照组,高剂量组的CAT水平明显高于对照组,高剂量组的GSH-Px水平明显高于对照组,差异均有统计学意义(P<0.05),但低剂量组的CAT水平和GSH-Px水平升高均不明显,差异均无统计学意义(P>0.05)。结论 扶芳藤总三萜可提高衰老小鼠抗氧化能力,具有抗衰老功效。

Effect and significance of total triterpenes of Euonymus fortunei (Turcz.) hand-mazz on the expression ofT-AOC, CAT, and GSH-Px in serum and tissues of mice with D-galactose induced senescence.

【Abstract】 Objective To explore the antioxidant effects of triterpenes from Euonymus fortunei (Turcz.)hand-mazz on D-galactose induced senescence in mice. Methods A total of 75 healthy and clean male Kunming micewere collected and subcutaneously injected with D-galactose to establish a model of aging mice. The mice were dividedinto positive drug group, model group, high and low total triterpenes of Euonymus fortunei (Turcz.) hand-mazz, and nor-mal control group by simple random sampling, with 15 mice in each group. After 21 days of continuous administrationin each group according to the preset schedule, the serum, heart, liver, and skeletal muscle tissues of mice were tested fortotal antioxidant capacity (T-AOC), catalase (CAT), and glutathione peroxidase (GSH-PX). Results Compared with thecontrol group, the serum T-AOC and GSH-Px levels of the mice in the high and low dose groups increased, and theT-AOC level in the low dose group was significantly higher than that in the control group (P<0.05); the T-AOC andGSH-Px levels in other groups were not significantly increased (P>0.05). Compared with the control group, the levels ofT-AOC, CAT, and GSH-Px in the heart tissue of mice in the high and low dose groups increased; the levels of T-AOC inthe high and low dose groups were significantly higher than those in the control group, and the CAT in the high dosegroup was significantly higher than that in the control group (P<0.05); the levels of T-AOC, CAT, and GSH-Px in othergroups were not significantly increased (P>0.05).
