      标题:基于网络药理学预测鸡屎藤治疗类风湿关节炎作用机制

      【摘要】 目的 运用网络药理学分析鸡屎藤治疗类风湿关节炎(RA)的类药活性成分和作用机制。方法 在各大传统中药数据库以及化合物信息数据库检索挖掘鸡屎藤活性成分,基于ADME原理筛选类药成分,利用虚拟对接平台预测筛选各类药成分靶蛋白,分析化合物-蛋白-疾病互作网络,确定鸡屎藤治疗类风湿关节炎的重要靶点,结合生物信息学手段、GO分析及KEGG信号通路富集,分析鸡屎藤治疗类风湿关节炎的类药活性成分和具体机制。结果 获得鸡屎藤类药成分11种,数据库挖掘及维恩分析获得疾病-化合物重叠靶基因62个,筛选出关键节点与Hub节点,富集分析显示鸡屎藤可能通过凋亡、炎症反应、血管翳生成等多个方面对多个靶点、多个通路产生调控作用。结论 通过网络药理学分析,鸡屎藤可能影响凋亡、炎症反应、血管翳生成等几条信号通路,影响RA进程中各种细胞功能,起到治疗类风湿关节炎的作用,为类风湿关节炎的研究提供了方向。

Prediction of the mechanism of action of Paederia scandens in the treatment of rheumatoid arthritis based onnetwork pharmacology.

【Abstract】 Objective To study the active constituents and mechanism of action of Paederia scandens in thetreatment of rheumatoid arthritis (RA) by network pharmacology analysis. Methods Paederia scandens active ingredi-ents were searched in the traditional Chinese medicine database and compound information database. The drug compo-nents were screened based on ADME (absorption, distribution, metabolism, excretion, ADME) principle. The virtualdocking platform was used to predict and screen the target proteins of various drug components, and the compound-pro-tein-disease interaction network was analyzed to determine the important target of Paederia scandens in the treatment ofRA. Combined with bioinformatics, GO analysis and KEGG signaling pathway enrichment, the active ingredients andspecific mechanisms of Paederia scandens in the treatment of RA was analyzed. Results Eleven Paederia scandens drugcomponents were obtained, 62 disease-compound overlapping target genes were obtained by database mining and Wienanalysis, and key nodes and hub nodes were screened. Enrichment analysis showed that Paederia scandens may regulatemultiple targets and pathways through apoptosis, inflammatory reaction and vasospasm. Conclusion Network pharma-cology analysis shows that Paederia scandens may affect several signaling pathways such as apoptosis, inflammatory re-sponse, and vasospasm, and play a role in the treatment of rheumatoid arthritis by affecting various cellular functions inthe RA, which provides direction for the study of rheumatoid arthritis.
