      标题:素馨护肝方对ANIT诱导的大鼠胆汁淤积肝损伤的保护作用

      【摘要】 目的 观察素馨护肝方对血清谷丙转氨酶(ANIT)诱导的大鼠胆汁淤积肝损伤血清氨酶及总胆红素(T-BIL)、直接胆红素(D-BIL)、间接胆红素(I-BIL)、总胆汁酸(TBA)、血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、γ-谷氨酰转移酶(γ-GGT)的影响,肝组织总超氧化物歧化酶(T-SOD)、丙二醛(MDA)含量变化及肝组织的病理改变,探讨该复方防治肝内胆汁瘀积肝损伤的保护作用。方法 SD雄性大鼠84只随机分为正常对照组、模型组、阳性药熊去氧胆酸组和素馨护肝方组,每组21只,各组按处理时间分为24 h、48 h、72 h三个亚组,每个亚组7只大鼠,给药组给予相应的药物,模型组和正常对照组给予生理盐水,连续灌胃4 d,实验第5天,除正常对照组外,其他各组一次性灌胃ANIT橄榄油,灌胃前12 h禁食不禁水,正常对照组灌胃等体积的橄榄油。造模4 h后各组给予相应的药物或生理盐水。之后分别于造模后24 h、48 h、72 h处死相应动物,处死前禁食不禁水12 h。空腹取血,收集肝脏标本。全自动生化分析仪检测血清T-BIL、D-BIL、I-BIL、TBA、ALT、AST、ALP、γ-GGT含量;按试剂盒操作检测肝组织中T-SOD和MDA含量及肝组织做病理学检测。结果 与模型组比较,素馨护肝方组和熊去氧胆酸组在24 h、48 h、72 h三个时间节点的血清T-BIL、D-BIL、I-BIL、TBA、ALT、AST、ALP、γ-GGT水平均明显低于模型组,差异均有统计学意义(P<0.05),与模型组比较,素馨护肝方组和熊去氧胆酸组在24 h、48 h及72 h三个时间节点均能显著升高肝组织匀浆中T-SOD含量,显著降低MDA水平,差异均有统计学意义(P<0.05);在不同时间点,上述指标在素馨护肝方组与熊去氧胆酸组之间比较差异无统计学意义(P>0.05)。HE染色显示,素馨护肝方组减少炎性细胞浸润范围和纤维细胞增生程度。结论 素馨护肝方能明显抑制肝内胆汁瘀积大鼠血清T-BIL、D-BIL、I-BIL、TBA、ALT、AST、ALP、γ-GGT水平,升高肝组织中T-SOD含量,降低其MDA水平,从而减轻肝细胞的损伤,表现出较好的肝损伤保护作用,这可能是其防治肝内胆汁瘀积肝损伤的重要机制之一。

Protective effect of Suxin Hugan Prescription on alpha-naphthylisothiocyanate-induced hepatic injury in ratswith cholestasis.

【Abstract】 Objective To observe the effects of Suxin Hugan Prescription on serum ammonia, total bilirubin(T-BIL), direct bilirubin (D-BIL), indirect bilirubin (I-BIL), total bile acid (TBA), serum alanine aminotransferase(ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), gamma-glutamyl transferase (gamma-GGGT),and total superoxide dismutase (T-SOD), and malondialdehyde (MDA), as well as pathological changes of liver tissue inrats with cholestasis induced by alpha-naphthylisothiocyanate (ANIT), and to explore the protective effect of SuxinHugan Prescription on hepatic injury. Methods Eighty-four SD male rats were randomly divided into four groups: nor-mal control group, model group, ursodeoxycholic acid group, Suxin Hugan Prescription group, with 21 rats in eachgroup. Each group was divided into three subgroups according to the treatment time: 24 h, 48 h and 72 h, with seven ratsin each subgroup. The ursodeoxycholic acid group and Suxin Hugan Prescription group were given corresponding drugs,and the model group and normal control group were given saline, all for 4 consecutive days. On the fifth day of the ex-periment, the normal control group was given the same volume of olive oil, and other groups were given ANIT olive oilonce (abstinence from food for 12 hours before gavage, but not water). After 4 hours, each group was given correspond-ing drugs or saline. The corresponding animals were executed 24, 48 and 72 hours after the establishment of the model,and the animals were fasting for 12 hours before the execution. Blood samples and liver samples were collected. SerumT-BIL, D-BIL, I-BIL, TBA, ALT, AST, ALP, and gamma-GGT were analyzed by automatic biochemical analyzer, livertissue T-SOD and MDA were detected, and pathological examination was performed. Results Compared with modelgroup, the levels of T-BIL, D-BIL, I-BIL, TBA, ALT, AST, ALP, and gamma-GGT in serum of Suxin Hugan Prescription基金项目:广东省中山市科技计划项目(编号:2017B1111)
