      标题:CPT1A促进肺癌转移的调控作用

      【摘要】 目的 研究肉毒碱棕榈酰基转移酶1A (CPT1A)在肺癌转移中的调控作用。方法 首先,利用实时定量PCR (qRT-PCR)与Western blot实验检测11对肺癌原发灶与转移灶组织中CPT1A分子的mRNA与蛋白表达,以明确CPT1A在肺癌转移过程中的表达是否发生改变;其次,用细胞划痕实验检测下调CPT1A表达对肺癌A549细胞迁移能力的影响;最后,用Transwell侵袭实验检测下调CPT1A表达对肺癌A549细胞侵袭能力的影响。结果 肺癌转移灶组织中CPT1A表达在mRNA与蛋白水平均明显高于原发灶组织[mRNA水平:(1.00±0.14)vs (1.83±0.75);蛋白水平:(1.00±0.26) vs (2.25±0.63)],差异均有统计学意义(P<0.05);与对照组细胞相比,下调CPT1A表达可显著抑制肺癌细胞的迁移能力[siCtrl vs siCPT1A-1 vs siCPT1A-2=(1.00±0.12) vs (0.39±0.04) vs (0.38±0.03)],差异均有统计学意义(P<0.05);与对照组相比,下调CPT1A表达可显著抑制肺癌细胞的侵袭能力[siCtrl vs siCPT1A vs siCPT1A-2=(23.00±3.00) vs (12.00±2.00) vs (13.00±1.00)],差异均有统计学意义(P<0.05)。结论 CPT1A在肺癌转移过程中表达显著上调;CPT1A可同时促进肺癌细胞的迁移与侵袭能力,提示CPT1A是肺癌治疗潜在的分子靶标。

Roles of CPT1A promoting lung cancer metastasis.

【Abstract】 Objective To investigate the regulation of carnitine palmitoyltransferase 1A (CPT1A) in lung can-cer metastasis. Methods Firstly, real-time quantitative PCR (qRT-PCR) and Western blot were used to detect the ex-pression of CPT1A mRNA and protein in 11 pairs of primary and metastatic tissues of lung cancer, and to determinewhether the expression of CPT1A in lung cancer metastasis changed. Secondly, scratch wound healing assay was used todetect the effect of down-regulation of CPT1A expression on the migration ability of lung cancer cells (A549 cell line).Finally, Transwell invasion assay was used to evaluate the effect of down-regulating CPT1A expression on the invasionability of lung cancer A549 cells. Results The expression level of CPT1A in lung cancer metastatic tissues was signifi-cantly higher than that in primary tissues: mRNA level: (1.00±0.14) vs (1.83±0.75); protein level: (1.00±0.26) vs (2.25±0.63); P<0.05. Compared with the control cells, down-regulation of CPT1A expression significantly suppressed the mi-gration ability of lung cancer cells: siCtrl vs siCPT1A-1 vs siCPT1A-2 = (1.00±0.12) vs (0.39±0.04) vs (0.38±0.03), P<0.05. Down-regulation of CPT1A expression also significantly inhibited the invasion abilities of lung cancer cellscompared with the control group: siCtrl vs siCPT1A-1 vs siCPT1A-2 = (23.00 ± 3.00) vs (12.00 ± 2.00) vs (13.00 ±1.00), P<0.05. Conclusion The expression levels of CPT1A is significantly up-regulated during the metastasis of lungcancer. CPT1A can simultaneously promote the migration and invasion of lung cancer cells, suggesting that CPT1A is apotential molecular target for the treatment of lung cancer.
