      标题:H19基因rs3741219多态性与口腔鳞状细胞癌的相关性

      【摘要】 目的 分析H19基因 rs3741219位点单核苷酸多态性与中国汉族人群口腔鳞状细胞癌(OSCC)的相关性,并分析吸烟和饮酒与OSCC发病风险的关系。方法 收集2016年7月至2017年11月期间在十堰市太和医院住院的119例OSCC患者(病例组),以114例无癌症病史且年龄和性别均匹配者为对照组。运用直接法分别对吸烟率和饮酒率进行性别校正后比较两组间差异。采用PCR-RFLP方法对两组受检者的 rs3741219位点进行基因型检测,运用χ2检验和单因素、多因素Logistic回归等分析其与OSCC发生的相关性。结果 病例组与对照组人群在年龄和性别构成方面均衡可比(P>0.05),但病例组患者的吸烟率(62.2%)和饮酒率(48.7%)较对照组的吸烟率(33.3%)和饮酒率(29.8%)显著增高,且通过性别进行率的标准化后的结果仍显示病例组高,差异均有统计学意义(P<0.01);关联研究结果显示,病例组患者的TT、TC和CC基因型频率为47.1%、45.4%和7.6%,对照组分别为41.2%、51.8%和7.0%,频率分布的组间比较差异均无统计学意义(P>0.05);多因素Logistic分析结果显示,病例组和对照组的TC和CC基因型的分布频率比较差异均无统计学意义(P>0.05),但分层分析结果显示,在年龄≥60岁和吸烟人群中,rs3741219位点 TC基因型携带者较 TT+CC基因型携带者会显著增加OSCC发病风险,差异有统计学意义(P<0.05)。结论 (1) Rs3741219位点多态性改变可能与中国汉族人群OSCC的发病有关;(2)长期吸烟和饮酒可能会增加OSCC的发病风险。

Association between rs3741219 polymorphism in H19 gene and oral squamous cell carcinoma.

【Abstract】 Objective To investigate the relationship between the rs3741219 polymorphism in H19 gene andthe susceptibility of oral squamous cell carcinoma (OSCC), and to analyze the association between tobacco and alcoholintake with the risk of OSCC. Methods A total of 119 OSCC cases (the OSCC group) and 114 age- and sex-matchedcancer-free controls (the control group) were collected from Taihe Hospital from July 2016 to November 2017. Thesmoking and drinking rates were gender-adjusted by the direct correction method and the difference between the twogroups was compared. PCR-RFLP method was used to genotype the rs3741219 polymorphism. The chi-square test andLogistic regression methods were used to analyze the association. Results There were no significant differences in ageand sex between the OSCC group and control group (P>0.05). However, the proportions of smokers and drinkers in theOSCC patients were both significantly higher than those in the control group (62.2% vs 33.3%, 48.7% vs 29.8%, respec-tively), and the gender-adjusted results also revealed similar trend (P<0.01). The frequencies of TT, TC, and CC geno-type were 47.1%, 45.4%, and 7.6% in the OSCC group, and 41.2%, 51.8%, and 7.0% in the control group, respectively,and there was no statistically significant difference between the groups (P>0.05). Multivariate Logistic analysis showedthat the distribution frequency of TC and CC genotypes was not significant different between the case group and the con-trol group (P>0.05). However, association study showed that the TC genotypes carriers of rs3741219 polymorphism hadsignificantly increased risk of OSCC than the TT+CC genotypes carriers among the age≥ 60 years and smokers in thestratified analysis, respectively (P<0.05). Conclusion (1) Rs3741219 polymorphism may be one susceptibility locusfor OSCC in the Chinese Han population. (2) Long-time tobacco and alcohol intake may increase the risk of OSCC.
